Submitted for the MAR15 Meeting of The American Physical Society Quantitative analysis of actin monomer funneling: how capping protein enhances actin filament growth and nucleation on biomimetic beads1 RUIZHE WANG, ANDERS CARLSSON, Washington University — Capping protein (CP) caps the growing ends of actin filaments and thereby halts their polymerization. However, CP is required for actin-based motility, and experiments by Akin and Mullins [1] have shown that CP also enhances the rate of filament nucleation. Proposed explanations for these phenomena include the Actin Funneling Hypothesis (AFH) [2], in which the presence of CP increases the free-actin concentration, and structural changes of the actin networks induced by increasing CP [1]. In this article, we provide a quantitative analysis of the AFH based on rate equations including actin nucleation and branching, polymerization and capping, plus monomer depletion near the surface of the bead. With two adjustable parameters, our simulation results accurately match several aspects of the results of Akin and Mullins [1]. We find that CP increases the local monomer concentration at the bead surface, but has a much smaller effect on the global free-actin concentration. The increased local monomer concentration gives rise to an enhanced rate of branching events and thus a larger number of actin filaments. [1] O Akin and R. D. Mullins. Cell 133.5 (2008): 841-851. Enhanced Depolymerization of Actin Filaments by ADF/Cofilin and Monomer Funneling by Capping Protein Cooperate to Accelerate Barbed-End Growth

A living cell's ability to assemble actin filaments in intracellular motile processes is directly dependent on the availability of polymerizable actin monomers, which feed polarized filament growth [1, 2]. Continued generation of the monomer pool by filament disassembly is therefore crucial. Disassemblers like actin depolymerizing factor (ADF)/cofilin and filament cappers like capping protein (...

Follow the Monomer

Capping proteins limit actin filament growth, but paradoxically increase actin-based cell motility. This has been attributed to funneling of actin monomers to the filament ends that remain uncapped. Using a reconstituted motility system, Akin and Mullins (2008) now demonstrate that filament capping increases Arp2/3-based nucleation and branching, rather than elevating the rate of filament elong...

Mechano-chemical feedbacks regulate actin mesh growth in lamellipodial protrusions.

During cell motion on a substratum, eukaryotic cells project sheetlike lamellipodia which contain a dynamically remodeling three-dimensional actin mesh. A number of regulatory proteins and subtle mechano-chemical couplings determine the lamellipodial protrusion dynamics. To study these processes, we constructed a microscopic physico-chemical computational model, which incorporates a number of f...

Actin filament branching and protrusion velocity in a simple 1D model of a motile cell.

We formulate and analyse a 1D model for the spatial distribution of actin density at the leading edge of a motile cell. The model incorporates nucleation, capping, growth and decay of actin filaments, as well as retrograde flow of the actin meshwork and known parameter values based on the literature. Using a simplified geometry, and reasonable assumptions about the biochemical processes, we der...

Effect of capping protein on the kinetics of actin polymerization.

Acanthamoeba capping protein increased the rate of actin polymerization from monomers with and without calcium. In the absence of calcium, capping protein also increased the critical concentration for polymerization. Various models were evaluated for their ability to predict the effect of capping protein on kinetic curves for actin polymerization under conditions where the critical concentratio...
